Characterization and performance assessment of the evolved strains from Actinobacillus succinogenes 130Z using Trolli streams is ongoing.
The resistance experiment had as aim to assess the performance of the adapted strains compared to the WT strain of the A. succinogenes 130Z. Four colonies from each agar plate (positive control, adapted strains, and the wild type (WT)) from day 12 (evolutionary adaptation experiment) were cultivated in 24 deep well-plates. Each well consisted of 5mL medium with 10 g/L glucose, 5 g/L yeast extract, 20.3 g/L K2HPO4.3H2O, 0.2 g/L MgCl2, 0.2 g/L CaCl2, 1 g/L NaCl, 9.6 g/L NaH2PO4.2H2O as nutrients, and 10 g/L NaHCO3.
Optical density (OD) is monitoring at 660 nm and dilutions to reach 103 cell/μL are performed to inoculate the 96 well-plates (4 plates for the positive control, 4 plates for the adapted strains and, 1 plate for the WT). The sugar concentration of trolli sample is adjusted to range from 20 to 120 g/L in the 96 well plate.
